    An endocytic-secretory cycle participates in Toxoplasma gondii in motility

    Apicomplexan parasites invade host cells in an active process involving their ability to move by gliding motility. While the acto-myosin system of the parasite plays a crucial role in the formation and release of attachment sites during this process, there are still open questions regarding the involvement of other mechanisms in parasite motility. In many eukaryotes, a secretory-endocytic cycle leads to the recycling of receptors (integrins), necessary to form attachment sites, regulation of surface area during motility, and generation of retrograde membrane flow. Here, we demonstrate that endocytosis operates during gliding motility in Toxoplasma gondii and appears to be crucial for the establishment of retrograde membrane flow, because inhibition of endocytosis blocks retrograde flow and motility. We demonstrate that extracellular parasites can efficiently incorporate exogenous material, such as labelled phospholipids, nanogold particles (NGPs), antibodies, and Concanavalin A (ConA). Using labelled phospholipids, we observed that the endocytic and secretory pathways of the parasite converge, and endocytosed lipids are subsequently secreted, demonstrating the operation of an endocytic-secretory cycle. Together our data consolidate previous findings, and we propose an additional model, working in parallel to the acto-myosin motor, that reconciles parasite motility with observations in other eukaryotes: an apicomplexan fountain-flow-model for parasite motility

    A highly dynamic F-actin network regulates transport and recycling of micronemes in Toxoplasma gondii vacuoles

    The obligate intracellular parasite Toxoplasma gondii replicates in an unusual process, described as internal budding. Multiple dausghter parasites are formed sequentially within a single mother cell, requiring replication and distribution of essential organelles such as micronemes. These organelles are thought to be formed de novo in the developing daughter cells. Using dual labelling of a microneme protein MIC2 and super-resolution microscopy, we show that micronemes are recycled from the mother to the forming daughter parasites using a highly dynamic F-actin network. While this recycling pathway is F-actin dependent, de novo synthesis of micronemes appears to be F-actin independent. The F-actin network connects individual parasites, supports long, multidirectional vesicular transport, and regulates transport, density and localisation of micronemal vesicles. The residual body acts as a storage and sorting station for these organelles. Our data describe an F-actin dependent mechanism in apicomplexans for transport and recycling of maternal organelles during intracellular development

    Parasites lacking the micronemal protein MIC2 are deficient in surface attachment and host cell egress, but remain virulent in vivo

    Background: Micronemal proteins of the thrombospondin-related anonymous protein (TRAP) family are believed to play essential roles during gliding motility and host cell invasion by apicomplexan parasites, and currently represent major vaccine candidates against Plasmodium falciparum, the causative agent of malaria. However, recent evidence suggests that they play multiple and different roles than previously assumed. Here, we analyse a null mutant for MIC2, the TRAP homolog in Toxoplasma gondii. Methods: We performed a careful analysis of parasite motility in a 3D-environment, attachment under shear stress conditions, host cell invasion and in vivo virulence. Results: We verified the role of MIC2 in efficient surface attachment, but were unable to identify any direct function of MIC2 in sustaining gliding motility or host cell invasion once initiated. Furthermore, we find that deletion of mic2 causes a slightly delayed infection in vivo, leading only to mild attenuation of virulence; like with wildtype parasites, inoculation with even low numbers of mic2 KO parasites causes lethal disease in mice. However, deletion of mic2 causes delayed host cell egress in vitro, possibly via disrupted signal transduction pathways. Conclusions: We confirm a critical role of MIC2 in parasite attachment to the surface, leading to reduced parasite motility and host cell invasion. However, MIC2 appears to not be critical for gliding motility or host cell invasion, since parasite speed during these processes is unaffected. Furthermore, deletion of MIC2 leads only to slight attenuation of the parasite

    Response of cetaceans to fluctuations of pelagic fish stocks and environmental conditions within the Celtic Sea ecosystem

    Inshore waters off the south coast of Ireland are an important foraging area for a range of cetacean species. Some of the main prey species of these cetaceans are herring and sprat, two economically valuable fish species in the region. The Celtic Sea herring stock suffered a marked decline in 2013. The present study aimed to investigate potential changes in the ecosystem associated to the herring decline and to determine the potential impacts on predators. Here we analyzed sightings information of common dolphins, fin, minke, humpback and unidentified whale species, acoustic data of herring and sprat, and a range of environmental variables mainly derived from satellites. Firstly, we characterized spatio-temporal patterns in the relative abundance of predator and prey species, and environmental variables, and compared periods before and since the herring decline. Since the 2013 herring decline, (i) the herring stock has mainly concentrated in south-eastern coastal waters and southern offshore Irish waters, (ii) sprat density has increased, (iii) chlorophyll concentration has decreased, (iv) sea surface temperature has risen, and (v) the euphotic layer has extended deeper. Secondly, we modelled the effects of prey density and environmental conditions on the relative abundance and distribution of cetaceans, as well as the effects of environmental conditions on prey density, between 2005-2018 by applying Hurdle Generalized Additive Models. The models for herring and sprat support the idea that these species have different environmental relationships, for example herring tended to be found in shallower waters than was the case for sprat. The presence and relative abundance of common dolphins were significantly affected by both environmental conditions and herring density, whereas whale species presence and relative abundance were found to be correlated with sea surface temperature and prey density. The model results suggest differences in prey choice among whale species. Understanding the dynamic relationships between predators, prey and the environment is important to inform an ecosystem-based approach to fisheries management

    DNA metabarcoding as a marine conservation and management tool: A circumpolar examination of fishery discards in the diet of threatened albatrosses

    Almost all of the world's fisheries overlap spatially and temporally with foraging seabirds, with impacts that range from food supplementation (through scavenging behind vessels), to resource competition and incidental mortality. The nature and extent of interactions between seabirds and fisheries vary, as does the level and efficacy of management and mitigation. Seabird dietary studies provide information on prey diversity and often identify species that are also caught in fisheries, providing evidence of linkages which can be used to improve ecosystem based management of fisheries. However, species identification of fish can be difficult with conventional dietary techniques. The black-browed albatross (Thalassarche melanophris) has a circumpolar distribution and has suffered major population declines due primarily to incidental mortality in fisheries. We use DNA metabarcoding of black-browed albatross scats to investigate their fish prey during the breeding season at six sites across their range, over two seasons. We identify the spatial and temporal diversity of fish in their diets and overlaps with fisheries operating in adjacent waters. Across all sites, 51 fish species from 33 families were identified, with 23 species contributing >10% of the proportion of samples or sequences at any site. There was extensive geographic variation but little inter-annual variability in fish species consumed. Several fish species that are not easily accessible to albatross, but are commercially harvested or by-caught, were detected in the albatross diet during the breeding season. This was particularly evident at the Falkland Islands and Iles Kerguelen where higher fishery catch amounts (or discard amounts where known) corresponded to higher occurrence of these species in diet samples. This study indicates ongoing interactions with fisheries through consumption of fishery discards, increasing the risk of seabird mortality. Breeding success was higher at sites where fisheries discards were detected in the diet, highlighting the need to minimize discarding to reduce impacts on the ecosystem. DNA metabarcoding provides a valuable non-invasive tool for assessing the fish prey of seabirds across broad geographic ranges. This provides an avenue for fishery resource managers to assess compliance of fisheries with discard policies and the level of interaction with scavenging seabirds

    Complementary and alternative medicine use in glioma patients in France

    PURPOSE Complementary and alternative medicine (CAM) use increases in cancer patients, including adult patients with diffuse gliomas. METHODS Questionnaires addressing CAM use were distributed to adult patients with gliomas of WHO grades II-IV and ECOG performance score of 0-2 during hospital visits and filled in anonymously. The study was conducted in nine centers in France from May 2017 to May 2018. Descriptive cohort analyses and comparative analyses according to gender, age, WHO grade, and recurrent versus newly diagnosed disease were conducted. RESULTS Two hundred twenty-seven questionnaires were collected; 135 patients (59%) were male. Median age was 48 years, 105 patients (46%) declared having glioblastoma, 99 patients (43%) declared having recurrent disease. Hundred-three patients (45%) had modified their alimentary habits after the glioma diagnosis. At the time of the questionnaire, 100 patients (44%) were on complementary treatment, mainly vitamins and food supplements, and 73 patients (32%) used alternative medicine approaches, mainly magnetism and acupuncture. In total, 154 patients (68%) declared using at least one of these approaches. Expenditures exceeding 100 € per month were reported by users in 14% for modification of alimentary habits, in 25% for complementary treatment, and in 18% for alternative medicines. All approaches were commonly considered as improving quality of life and experienced as efficient, notably those associated with more expenditures. CONCLUSIONS CAM are frequently used by glioma patients in France. Underlying needs and expectations, as well as potential interactions with tumor-specific treatments, and financial and quality of life burden, should be discussed with patients and caregivers

    Etude de la réponse à la sélection massale paysanne sur les variétés de maïs population

    Depuis une vingtaine d’années, les paysan·ne·s de Dordogne s’intéressent et travaillent sur la sélection de variétés de maïs population. Dans le cadre du projet COVALIENCE, les paysan·ne·s ont souhaité étudier des méthodes de sélection simples, afin de déterminer si celles- ci pouvaient permettre l’augmentation du rendement grain de leurs variétés. Ainsi, il a été constaté que la création d’un différentiel de sélection était possible grâce à l’utilisation d’un protocole de sélection simple, basé sur le poids épi. Cependant, l’utilisation d’un tel protocole allonge l’intervalle de floraison mâle/femelle des variétés, augmentant par conséquent leur vulnérabilité à la sécheresse. La castration des pieds mâles tardifs uniquement ne semble pas compenser cet effet. Les méthodes de sélection sont donc à adapter à chaque situation et peuvent être agrémentées d’autres éléments afin de répondre aux objectifs de chacun. Les conclusions de cette étude orientent les futurs travaux d’AgroBio Périgord vers des sélections basées sur les floraisons et la protandrie

    WORKSHOP ON GUIDELINES FOR MANAGEMENT STRATEGY EVALUATIONS (WKGMSE2)

    The purpose of the meeting was to bring up to date the methodologies and technical specifications that should be incorporated in Management Strategy Evaluation (MSE) work in ICES. The workshop was tasked with reviewing recent methodological and practical MSE work conducted in ICES and around the world, as well as the guidelines provided by the 2013 ICES Workshop on Guidelines for Management Strategy Evaluations (WKGMSE). The Terms of Reference indicated that the revision should include all aspects involved in MSE, while paying specific attention to several issues that had been identified through ICES practice. The Terms of Reference also requested WKGMSE 2 to consider how best to disseminate the guidelines to experts within the ICES community and the need for training courses. The workshop addressed all its Terms of Reference. The main results of the workshop are the revised MSE guidelines, as well as recommendations in relation to the ICES criterion for defining a management strategy as precautionary and in relation to the evaluation and advice on rebuilding strategies.publishedVersio
